Introduction
기초 연구와 산업 생산에서 발견되는 mycoplasma contamination은 cell line의 87%를 감염시킬 정도로 심각한 문제입니다. Bacteria, fungal contamination과 달리 cell culture 상태에서 mycoplasma contamination은 시각, 후각으로 알기 어렵습니다. 일반적으로 1 x 10e6 ~ 1 x 10e8 cfu/ml 수준으로 mycoplasma가 cultured cell에 존재하지만, inverted microscope (도립현미경)으로 볼 수 없습니다. Mycoplasma infection은 진핵세포의 성장, 모양을 변형시키며 cell function에도 영향을 끼치기 때문에 실험 결과의 variation이 심합니다. 따라서 주기적으로 mycoplasma detection kit를 활용하여 mycoplasma 유무를 확인하는 것이 중요합니다.
PlasmoTest™ - 세포를 이용한 발색반응 제품
PlasmoTest™ 는 cell-based colorimetric assay 제품이며, mycoplasma를 인식하는 Toll-like receptor 2 (TLR2)의 능력을 활용한 제품입니다. Mycoplasma가 culture 배지에 존재할 경우 HEK-Blue™-2 cell 표면의 TLR2는 activation되며, 이는 NF-κB와 AP-1을 활성화 시킵니다. 이들 transcription factor들이 최종적으로 SEAP (secreted embryonic alkaline phosphatase)의 분비를 유도하여 HEKBlue™ Detection이 포함되어 있는 배지에서 purple/blue로 색변화가 일어납니다. 따라서 mycoplasma 감염 여부를 육안 또는 OD 620-655 nm에서 확인할 수 있습니다.

[Fig 2.] PlasmoTest™ 사용방법
- Mycoplasma test를 원하는 cell culture supernatant 500 μl collection
- 100°C, 15 분간 열처리
- HEK-Blue™ water 50 ml에 HEK-Blue™ Detection을 녹여서 준비 (사용전 filtering 진행)
- 열처리한 sample을 50 μl씩 96-well plate로 transfer
- 제품에 포함되어 있는 control도 50 μl씩 96-well plate로 transfer
- Prewarmed HEK-Blue™ Detection medium에 HEK-Blue™-2 cell suspension 준비 (남은 HEK-Blue™-2 cell은 cell banking 후 최대 30 passage까지 사용 가능)
- Cell suspension을 well 당 200 μl (~50,000 cells) 첨가
- Plate를 37 °C, CO2 incubator에서 overnight (16-24 hours)
- Mycoplasma 존재 여부는 육안으로 또는 OD 620-655 nm에서 발색반응 확인 가능
