1. 제품 Overview
Glucose Uptake Assay Kit (Colorimetric) (ab136955)는 다양한 세포 유형에서 10 pmol/well의 낮은 포도당 흡수를 감지할 수 있는 매우 민감하고 사용하기 쉬운 비방사성(non-radioactive) 분석 키트입니다.
2-데옥시글루코스(2-DG)는 포도당과 구조적 유사성 때문에 포도당 섭취 분석 프로토콜에 사용됩니다. 2-DG는 포도당 수송체에 의해 흡수되어 2-DG-6-포스페이트(2-DG6P)로 대사됩니다. 2-DG6P는 더 이상 대사될 수 없으므로 세포 내에 축적됩니다. 축적된 2-DG6P는 세포에 의한 2-DG(또는 포도당) 흡수에 정비례합니다. 이 분석에서, 2-DG6P는 NADPH를 생성하기 위해 산화되며, 그 수준은 enzymatic recycling amplification reaction에 의해 결정될 수 있습니다.

3. Glucose Uptake Assay protocol overview
- 실험 설정에 따라 적절한 glucose starvation / uptake stimulation 하여 세포를 준비합니다.
- 2-DG를 세포에 첨가하고 37℃에서 20분 동안 배양합니다.
- PBS로 세포를 세척하여 세포 밖 2-DG를 제거합니다.
- Extraction buffer를 이용하여 반복적인 파이펫팅으로 세포를 용해합니다.
- 용해물을 동결/해동하기 위하여 85℃에서 40분 간 가열합니다.
- 얼음에서 5분 간 식힙니다.
- Neutralizing buffer를 추가하고 Spin 시킨 후 상층액을 새 tube로 옮깁니다.
- Well에 상층액 및 standard를 분주합니다.
- Reaction mix A를 첨가하고 37℃에서 1시간 동안 incubation 합니다.
- Extraction buffer를 추가하고 40분 동안 90℃로 가열합니다.
- 얼음 위에서 5분 동안 식힌 후 neutralizing buffer를 추가합니다.
- Reaction mix B 추가
- 37℃에서 kinetic mode의 microplate reader에서 2-3분마다 분석합니다.

- 96 well plate with clear flat bottom
- KRPH(Krebs-Ringer-Phosphate-Hepes) buffer
